We have identified a new human immunodeficiency virus in a Cameroonian woman. It is closely related to gorilla simian immunodeficiency virus (SIVgor) and shows no evidence of recombination with other HIV-1 lineages. This new virus seems to be the prototype of a new HIV-1 lineage that is distinct from HIV-1 groups M, N and O. We propose to designate it HIV-1 group P.

Den gode nyheten for kvinnen, er at hun er symptomfri uten behandling. Den dårlige nyheten for KTere er, for å sitere fra det fullstendige paperet at når forskerne lette «for a divergent virus by using a nonspecific extra-long RT-PCR method» så lyktes de:

We successfully amplified the viral genome with this approach, allowing us to fully sequence it. …Evolutionary analysis of the near-complete genome sequence (Supplementary Methods) shows that the RBF168 strain is most closely related to SIVgor (Fig. 1a and Supplementary Fig. 3), and similarity plotting confirms that this relationship is maintained in all regions of the genome

Forbløffende presis kunnskap for et ikke-eksisterende fenomen.

Here we report the structure of an entire HIV-1 genome at single nucleotide resolution using SHAPE, a high-throughput RNA analysis technology. The genome encodes protein structure at two levels. In addition to the correspondence between RNA and protein primary sequences, a correlation exists between high levels of RNA structure and sequences that encode inter-domain loops in HIV proteins. This correlation suggests that RNA structure modulates ribosome elongation to promote native protein folding. Some simple genome elements previously shown to be important, including the ribosomal gag-pol frameshift stem-loop, are components of larger RNA motifs. We also identify organizational principles for unstructured RNA regions, including splice site acceptors and hypervariable regions. These results emphasize that the HIV-1 genome and, potentially, many coding RNAs are punctuated by previously unrecognized regulatory motifs and that extensive RNA structure constitutes an important component of the genetic code.

Kanskje feirer jeg «bloggbursdag» in absentia – i morgen feirer bloggen to år. Med 827 innlegg.)
